Several team have demonstrated inflammation as a key player in physiology and how an altered inflammatory response lead to pathologies. Chronic inflammation is installed in many disease such as cardiovascular disease. Ageing is also considered as inflammatory state. Cardiovascular disease is the leading cause of death in the world, and their frequency increased with age. This project aims to better understand the interaction of circulatory cells (platelet) with their environment (endothelium), in a context of cardiovascular disease in extreme-aged people. We will adopt a multidisciplinary approach with _in vitro, ex vivo_ and in _vivo _models, using different techniques as flow cytometry, microscopy, biochemical, molecular biology etc.Plusieurs équipes ont montré que l’inflammation jouait un rôle clé en physiologie et comment une réponse altérée à la résolution de l’inflammation peut conduire à des pathologies aigues et/ou chronique. L’inflammation chronique peut se traduire en différentes pathologies dont les maladies cardiovasculaires. Le vieillissement est aussi considéré comme un état inflammatoire. Dans le monde, les maladies cardiovasculaires sont la principale cause de décès. Le vieillissement de la population mène aussi à une augmentation de ces pathologies cardiovasculaires. Ce projet de thèse de science vise à mieux comprendre les interactions entre les cellules circulantes (plaquettes) avec leur environnement (notamment les cellules endothéliales) dans un contexte de pathologie cardiovasculaire chez des personnes d’âge extrême. Nous adopterons une approche multidisciplinaire avec des modèles _in vitro, ex vivo_ et _in vivo_, en utilisant des techniques de culture cellulaire, de cytométrie en flux, microscopie confocale, biochimique, biologie moléculaire etc.

The host laboratory DVH (laboratoire Dysfonction Vasculaire et Hémostase for Vascular Dysfunction and Hemostasis) is a part of INSERM unit Sainbiose.

DVH team has a strong expertise in vascular dysfunction and in hemostasis. The team is worldwide recognized. The DVH team grouped scientist, clinical scientist, postdoctoral fellow, PhD student, undergrad student and permanent staff. Le laboratoire d’accueil, DVH (laboratoire Dysfonction Vasculaire et Hémostase) fait partie de l’unité INSERM SAINBIOSE 1059.

Le laboratoire DVH à une forte expertise en dysfonction vasculaire mais aussi en hémostase, et est reconnu mondialement. Ce laboratoire est composé de chercheurs permanents, de clinicien scientifique, d’étudiant post doctoraux, d’étudiant en thèse, d’ingénieur et de techniciens. L’unité, le laboratoire est situé sur un campus riche en plateforme scientifique.

La thèse sera réalisée plus précisément dans l’équipe plaquette et inflammation au sein de DVH sous l’encadrement des Drs Anne Claire Duchez et Fabrice Cognasse

The PhD student will experimentally conduct the various aspects of the project, and will analyze the results. She/he will have to present her/his results weekly to her/his supervisors, during lab meeting and at scientific congresses.

**Required Skills**:Applicants must have:

- Master’s degree in research or equivalent in Biology, Biomedical Sciences
- Knowledge of immunology, hematology and cell biology
- Project relevant skills (but not essential) include cell biology, microscopy, immunofluorescence, immunohistochemistry, molecular biology, statistical analysis and data analysis
