Investigating the CIC::DUX4 Protein Interactions and Small Molecule Inhibition

The
Genetics, Epigenetics and Biology of Sarcoma
research team is seeking a highly motivated and creative post-doctoral fellow to investigate the interactome of the CIC::DUX4 oncoprotein. This project aims to elucidate the functional consequences of its interactions in CIC-Fused sarcoma and to identify small molecules capable of disrupting them, with potential therapeutic implications.

Research Project:

CIC-fused sarcomas are aggressive cancers with limited treatment options. We have gathered evidences suggesting that some specific interactors of CIC::DUX4 play critical roles in this sarcoma pathogenesis. In the frame of an INCa PLBIO funding, this project will focus on:

  • Characterizing
    the CIC::DUX4 interactions using advanced biochemical and cell biology techniques.
  • Deciphering
    the downstream signaling pathways affected by these interactions.
  • Validating
    small molecules to inhibit these interactions, with the goal of developing novel therapeutic strategies.

This project involves collaboration with biologists, chemists, physicists and bioinformaticians.

Skills and Expertise Required:

We are looking for candidates with a strong background in
molecular biology, biochemistry and cancer biology
, and experience in the following areas:

Molecular Biology:
qRT-PCR, Western blotting, co-immunoprecipitation, ChIP-seq, Cut&Tag, AID2 CRISPR/Cas9 gene editing.

Biochemistry:
Protein-protein interaction assays (e.g., GST pull-down, HTRF, RIME)

Cell Biology:
Cell culture, immunofluorescence, proximity ligation assays, functional cell-based assays

Drug Discovery:
Dose-response assays

Data Analysis:
Proficiency in statistical and sequencing analysis software (GraphPad, R, Python)

Additional Assets:

  • Experience in sarcoma research or related cancer models
  • Familiarity with epigenetic mechanisms and chromatin biology
  • Experience with zebrafish models of cancer
  • Strong publication record in peer-reviewed journals
  • Excellent communication skills and ability to work in a collaborative environment
