Laboratory

Our research team is interested in the spatio-temporal regulation of the mechanisms that govern genome stability in response to perturbations to the DNA replication process. Flaws in the DNA replication process, known as replication stress (RS), is a source of genome instability contributing to genomic disorders including cancer1. The causes of replication stress are many and varied but ultimately result in stressed replication forks that are fragile DNA structures prone to chromosomal breakage, rearrangement and mutation2. Our team aims to understand the spatial and temporal organization of molecular circuits that prevent stressed forks from being converted into pathological DNA structures. We investigate how recombination, repair, chromatin and RNA-based processes prevent replication stress-induced genome instability within the sub-nuclear architecture of the genome, using the yeast S. pombe and human cells as model systems. We combine genetic, cellular, genomic and molecular approaches to tackle these questions. Our team is based at the Institut Curie, on the Orsay site, about 20 km south of Paris. Our team is looking for a highly motivated post-doctoral researcher keen to understand how nuclear organization contributes to maintaining genome stability in response to replication perturbations. The candidate will join a research team of around 8 people, made up of PhD students, post-docs, engineers and researchers, to carry out his or her research in a collaborative and interactive spirit, while encouraging the taking of responsibility and initiative.

The project

The research project aims to decipher how nuclear architecture contributes to the maintenance of genome stability in response to replication perturbations. Our team has identified the nuclear pore complex as a molecular hub for promoting replication fork repair in the yeast S. pombe. The research project aims to understand the spatial specification of replication fork integrity pathways. The aim is to determine whether all nuclear compartments have the same capacity to preserve genome stability in response to replication perturbations, and to identify the underlying molecular mechanisms. The project requires the implementation of innovative proteomic and genomic approaches, and is part of a consortium (Space-ForkIn) funded by the ANR.

Training and Skills required

Candidate should hold a PhD in biology, cellular biology, or related areas. Fresh PhD graduate or with up to 3 years of postdoctoral experience. French is a plus.
