**Missions**:
La pro-opiomélanocortine (POMC) est un précurseur polypeptidique d'hormones et de neuropeptides (c'est-à-dire l'ACTH, l'a-MSH et la b-endorphine) aux fonctions multiples. La POMC exprimée dans le système nerveux central est un régulateur clé de l'homéostasie énergétique et du poids corporel chez les mammifères. Les variantes pathogènes bialléliques de la POMC provoquent une obésité sévère et précoce associée à une insuffisance hormonale de la glande surrénale et à des épisodes d'hypoglycémies. Il convient de noter que des peptides dérivés de la POMC ont également été détectés dans le pancréas endocrinien de mammifères, y compris les humains, suggérant une biosynthèse locale. Bien qu’un lien entre les peptides dérivés de la POMC, les cellules bêta pancréatiques et la sécrétion d’insuline aient étés rapportés, les mécanismes sous-jacents restent insaisissables. Nous émettons l'hypothèse que la POMC pancréatique pourrait réguler les îlots de Langerhans et en particulier les cellules bêta pancréatiques pour moduler la sécrétion d'insuline de manière paracrine ou autocrine. Nous proposons donc de caractériser l'expression du précurseur de la POMC et des peptides dérivés, des enzymes de transformation et des récepteurs dans différents modèles humains et murins in vitro et in vivo pour étudier le rôle de la POMC pancréatique sur la sécrétion d'insuline.

**Activités**:

- Expérimentation animale, incluant la dissection, l’isolation des îlots, les analyses métaboliques in vivo chez le modèle murin etc ;
- Biochimie, biologie moléculaire et cellulaire ;
- Analyses histologiques et immunohistologiques ;
- Analyses de l’expression des gènes ;
- Participation active dans tous les réunions de groupes.

Le post-doctorant sera responsable de la préparation, de l’interprétation et de la diffusion des résultats, y compris la rédaction d’articles de recherche et de présentations lors de conférences.

**Compétences**:

- Nous recherchons un expert en physiologie et métabolisme de la souris, plus particulièrement dans le domaine de la recherche sur le diabète.
- Doctorat avec une solide expérience en physiologie et métabolisme de la souris ;
- Compétences pour le travail en équipe et le travail indépendant ;
- Contexte de l'analyse du métabolisme in vivo chez les rongeurs ;
- Formation en biologie cellulaire, biologie moléculaire et physiologie, analyse tissulaire (histologie, IF);
- Expérience en analyse de l'expression génique ;
- Maîtrise de la langue anglaise (écrite et parlée) ;
