The Mondor Institute for Biomedical Research (IMRB, U955 Inserm - Université Paris Est Créteil, UPEC) is one of the main biomedical research centres in the eastern part of the Paris region with national and international influence. The research teams develop high-level translational research in a wide variety of fields in direct connection with healthcare services and a large number of patient cohorts. The research teams are structured into 3 departments: “Virus, Immunity, Cancer” (VIC, 5 teams); “Neurosciences and Psychiatry” (ESPRY, 3 teams), and “Physiopathology of Cardiovascular and Respiratory Diseases, Development and Senescence” (PHYDES, 6 teams).
**Description**:
- General context and positioning of the project_
- Obesity induced by nutritional stress such as high fat diet (HFD), is a major risk factor leading to several metabolic dysfunction such as type 2 diabetes and cardiovascular disease. Accumulation of fat during obesity is associated with chronic inflammation and hyperglycemia contributing to the development of insulin resistance. In addition to these well-known effects on metabolism, obesity also impairs the skeletal muscle, promoting a decline in muscle mass, quality and contractile function. The accumulation of fat surrounding the muscle over time likely contributes to obesity-mediated muscle atrophy and dysfunction.-
- Objectives_
- The major objective of this project is to understand the mechanisms by which CO can protect and maintain skeletal muscle structure and function during HFD. Our focus will be to assess muscle function, histological muscle parameters and atrophy markers in obese mice treated with CORM-401.- Scientific/technical program._
- Experiments will be performed using mice treated with either a standard diet (SD), a HFD or HFD in combination with CORM-401 over 14 weeks. Body weight will be monitored weekly and metabolic parameters such as glucose tolerance test (GTT) and insulin tolerance test (ITT) will be assessed. The effect of CO on muscle function during HFD will be explored by performing functional skeletal muscle tests and evaluating muscle contractility in the group under analysis. Histological analysis and skeletal muscle markers (fiber size and type, myosin, stem cell activity and others) will be assessed by immunostaining in samples collected from mice on HFD and HFD+CORM-401 and will be compared to SD mice.- Finally, we will investigate whether CO treatment in HFD mice modulates molecular markers of muscle atrophy and regeneration using qPCR and western blot techniques.**Profil**:
