- Dysregulated signaling pathways controlling cell metabolism can lead to uncontrolled cell proliferation and cancer. Accordingly, mutations of the effectors involved in the Pi3k-Akt-mTOR signaling pathway often result in T cell acute lymphoblastic leukemias (T-ALL). In this frame, we have identified a transcription factor (TF) that is a substrate of AKT and mTor kinases and which controls T cell metabolism. The proposed project will consist in investigating the role of this TF in T-cell leukemia, and determine whether its targeting could be used to control cancer progression.- The goal of the project is to investigate at the molecular and cellular level, the role of a new transcription factor in the development and progression of T-ALL in mouse models and humans.-
- In parallel, He/She will assess the importance of the TF in human T-ALL. First, He/She will compare protein expression, phosphorylation and cellular localization (cytoplasmic or nuclear) of the TF between Pten-sufficient and Pten-deficient T-ALL cell lines; importantly, these analyses will be extended to primary human samples. The generation of patient-derived xenografts (PDX) in which the TF is inactivated, is also planned to determine its impact in primary T-ALL in vivo.**Nature du financement**:
