**Postdoctoral Position on Gliovascular Unit of Glioblastoma**
Team leader and Co-funder of the Cancer and Cognition Platform

We are looking for a motivated and expert post-doc to join our team within the genetic and translational expert research Cancer and Brain Genomics INSERM unit, to work on the
GlioVasc PAIR Brain Tumor project (7 French teams). The global aim of the whole program is to elucidate the key features of the gliovascular unit and its pivotal role in glioblastoma (GB) recurrence and treatment resistance. We propose to explore the dynamic trajectories of the GB and stromal cells within the gliovascular unit from human GB samples and their molecular underpinnings during the Stupp protocol. Our NeuroGlio team will map the gliovascular unit heterogeneity phenotypically (organoids) and at the single cell level in GB patients and carry some gene targets validation in GB resection preclinical models.

**Proposed approaches**:Organoids (GBO) and gliovascular units (GBV) have already been implemented in the team from GB patient samples and from murine GB xenografts, and have been verified for maintenance of intra-tumor heterogenous cell composition and proliferation status. Some selected samples will be prepared for RNAseq/ATACseq analyses through the facilities of the Institute for Research and Innovation in Biomedicine of the Rouen Health
Institute site. Patient derived cell lines generated will also be generated to compare with gliovascular single-cell transcriptomics and epigenomics data of GB samples and for validation studies. In preclinical models of GB resection, GB heterogeneity and microenvironmental cells organization and composition will be systematically characterized by 2D and 3D imaging
(iDISCO and CLARITY) through local expert facilities (HeRacLeS) of Rouen.

**Team’s research programs**:We bring into play our competences in the field of
Neuroscience and Neuro-oncology in a very expert environment in the genetic predisposition of tumors, neuronal and neurodevelopmental pathologies. Our team aims to characterize new therapeutic targets in the context of brain tumors and to evaluate the brain sequelae related to cancer and cancer treatments, between therapeutic efficacy and preservation of brain functions. The strength lies in our expertise in molecular and cellular biology of migration and invasion of glioma cells and in the design and validation of dynamic cellular imaging tools. We also set up behavioral mouse models with cancer and after resection microsurgery, and during conventional treatments or immunotherapy. Neuroscience tools
(electrophysiology/optogenetics) are developed on organotypic GB slices and _in vivo_ GB mouse models. Then, clinical protocol and methodologies to establish GB organoids from patients and cerebroids from human iPS cells are currently leveraged in the team.

Imaris software, human scRNAseq analyses and in _in vivo_ preclinical models of GB growth.
Experience in bioinformatics would be much appreciated. He/she would be fluent in both spoken and written English.
