Myotendinous junction (MTJ) is a mechanical buffer of the skeletal muscle cell and prone site to injury upon exercise. Our lab recently developed a new tool to manipulate the MTJ domain and revealed its local translational program as well as how it rewires after exercise. However, the molecular mechanism by which this special translation program is achieved remains unknown. The student will take both candidate and unbiased approaches to address this problem.
- Use mouse genetics to generate mutants of candidate RNA binding proteins and dissect their phenotype.
- Investigate the local translatome upon mutating RNA binding proteins and identify their binding partners.
- Biochemical pull-down identification of candidate factors using affinity tag fused UTRs
- Presentation of the work in lab meetings and conferences.
- Supervising Master students.
Specific Requirements or Constraints :
- Mouse genetics skills: maintenance, genotyping, histological analyses, and imaging
- Physiological tests: various assays for phenotyping muscle function
- Biochemical skills for mass-spectrometry, NGS etc.
