Analysis of Human Normal Thyroid Organoids Response to Ionizing Radiation Specialist

Mission

Scientific Context

Thyroid exposure to ionizing radiation during childhood is a recognized risk factor for the development of cancers. However, the risk decreases with the dose, and statistical epidemiological methods are no longer applicable. To address this knowledge gap, we are developing an organoid model derived from human normal thyroid tissue, reproducing the 3D organization of thyroid follicles. The candidate will be involved in the analysis of the organoids' response to low doses of ionizing radiation.

Work Context and Objectives

We have culture conditions that permit to maintain follicular 3D structure, which synthesize the thyroid hormone thyroxine in the lumen. The candidate will be responsible for developing protocols focused on the analysis of the oxidative status of the organoids after exposure, including exploration of key signaling pathways, mitochondrial function, and reactive oxygen species production. This will be achieved in collaboration with the laboratory of T. Pourcher.

Long-Term Maintenance of Thyrospheres

The candidate will also be involved in the long-term maintenance of thyrospheres in culture, with the objective of following the response to ionizing radiation over the longest culture period. This will enable the analysis of possibly the early steps of carcinogenesis. The culture process should maintain the organoids' 3D structure and physiology over time.

Required Profile

Applicants should have a PhD in molecular and cellular biology. A good experience in cell culture and knowledge of the molecular mechanisms of the cellular response to genotoxic stress will be appreciated. The selected candidate will be encouraged to present the findings of the project at scientific conferences and to administrative authorities.
